dys-1::gfp VIII was expressed in the body wall muscles, the vulva muscles, the head muscles and the pharyngeal muscles. Staining was observed from 3-fold embryo to adulthood. No staining was detected in non-muscle cells. Other constructs containing shorter 3' sequences gave no or very weak signal.

qui-1::GFP was visible in the processes of the nerve ring and in several neurons of the lateral and ventral ganglia, including the avoidance sensory neurons ASH and ADL. GFP expression was not detected in ASK sensory neurons. Four additional neurons in the retrovesicular ganglion and two neurons in the lumbar ganglion, PVQ and the sensory neuron PHB, also express GFP. This expression pattern partially overlaps but is not completely coincident with that obtained with a much shorter construct in which GFP was fused to the second exon, and the construct was thus missing part of exon 2 and the last 17 introns and exons of the gene (not shown). Expression from this shorter construct included neither ASH nor PHB, indicating that important regulatory elements are present within the coding region of qui-1.

A shorter promoter with 2.8 kb of regulatory sequence upstream of the first exon of the d isoform (rfip-1p_d::gfp) had much more restricted expression than Expr12545 being found in the pharynx, the excretory canal, many head neurons, and a few tail neurons. Of note for this study, the rfip-1p_d::gfp reporter was expressed in the PLM, but not the ALM, neurons.

casy-1b and casy-1c showed co-localization with both cholinergic and GABAergic neuronal markers. casy-1b and casy-1c show a more limited expression than casy-1a in the head neurons. However, both casy-1b and casy-1c are strongly expressed in the VNC motor neurons, which is not seen with casy-1a. No expression was observed in the gonadal sheath for the shorter isoforms.

ptps-1::GFP transgenics with the shorter upstream sequence resembled the cat-4 expression pattern, with expression in some epidermal cells, and a few serotonergic neurons. The ptps-1 reporters, however, were also observed in additional cells (including anterior intestine, VC4, 5 (weak) and HSN, plus lateral seam cells), and were not expressed in other epidermal cells or in all 5HT and DA neurons, suggesting these reporters may lack important positive or negative regulatory elements.

A 6.7-kb fusion (LOV-1::GFP1) and a 15.4-kb fusion lacking only 58 amino acids (LOV-1::GFP5) direct expression in male-specific sensory neurons. Shorter versions of lov-1::gfp1 are not expressed in the same set of male-specific neurons, nor do they act as dominant negatives. lov-1::gfp1 and lov-1::gfp5 are expressed in male-sensory neurons, specifically the CEM neurons, the hook neuron HOB, and the sensory ray neurons. Expression begins during late L4 lethargus and peaks in the adult male.

The earliest detectable expression of UNC-115::GFP was in neurons and epidermis at about 300 min postfertilization, when the embryo begins to elongate, and axons begin to grow. UNC-115::GFP and the shorter fusion GFPS was expressed in most or all neurons throughout development. UNC-115::GFP expression was also observed in non neuronal cells, including the epidermal syncytium hyp7, the head and tail epidermal cells, the excretory canal cell, the pharynx, and the developing vulva, but not in the lateral epidermal seam cells or the ventral epidermal P cells.

GFP analysis of the transformed animals revealed that rcn-1 is expressed from late embryonic stages to adulthood in diverse tissues, including lateral hypodermal cells, marginal cells of the pharynx, vulva epithelial cells, ventral and dorsal nerve cords and commissures, and various neurons in the anterior and posterior portions of the worm. Male C. elegans displayed rcn-1 expression in male tail structures including the diagonal muscles, sensory rays, and spicules. GFP expression analysis of a shorter 1.6 kb 50 upstream fragment of rcn-1 revealed vulva muscle expression in addition to the aforementioned expression patterns.
